    Magnetic nanoparticles coated with carboxylate-terminated carbosilane dendrons as a reusable and green approach to extract/purify proteins

    Extraction/purification of proteins, at both analytical and industrial levels, is a limiting step that usually requires the use of organic solvents and involves tedious work and a high cost. This work proposes a more sustainable alternative based on the use of magnetic nanoparticles (MNPs) coated with carboxylate-terminated carbosilane dendrons. MNPs coated with first- and second-generation carbosilane dendrons and bare MNPs were employed for the extraction of proteins with different molecular weights and charges. Interaction of proteins with MNPs significantly varied with the pH, the protein, and the dendron generation (different sizes and number of charges in the periphery). Optimal dendron:protein molar ratios and suitable conditions for disrupting interactions after protein extraction were also researched. Second-generation dendron-coated MNPs showed 100% retention capability for all proteins when using acidic conditions. They were reused without losing magnetism or interaction capacity after a disruption of protein-dendron interactions with 0.2% SDS at 100 degrees C for 10 min.     El juego posee un gran potencial educativo y puede convertirse en una excelente herramienta para fomentar el desarrollo de los valores cívico-democráticos y la participación ciudadana desde edades tempranas. Partiendo de esta premisa, se desarrolla la iniciativa CHILDTIZENS, un proyecto de I+D cofinanciado por el Instituto Valenciano de Competitividad Empresarial (IVACE) de la Generalitat Valenciana y la Unión Europea mediante el Fondo Europeo de Desarrollo Regional. Cuyo objetivo es partir del valor educativo del juego para favorecer la concienciación y promoción de valores cívicos entre los niños y niñas. El proyecto contempla distintas fases que abarcan desde la revisión de la literatura académica reciente, hasta un estudio de mercado con una muestra inicial de 500 juguetes y juegos para valorar sus posibilidades de concienciación y promoción de actitudes cívicas. Fruto de este trabajo se ha desarrollado el CHILDTIZENS Participation Toolkit, un set de materiales lúdico-educativos destinados a la promoción de la participación ciudadana y la transformación optimizadora desde la infancia, centrado en 3 dimensiones básicas emergidas de la investigación: la accesibilidad, la sostenibilidad y la igualdad de género. Este set posibilita la transferencia de resultados de la investigación contribuyendo a avanzar hacia una sociedad más justa, cohesionada y comprometida.The game is a tool with a great educational potential that can promote the development of civic-democratic values and the citizen participation from an early age. Based on this premise, it is been carrying out the CHILDTIZENS initiative, an R&D project co-financed by the Valencian Institute of Business Competitiveness (IVACE) of the Generalitat Valenciana and the European Union through the European Regional Development Fund. Whose objective is to use the educational value of the game to favour awareness and promotion of civic values among little boys and girls. The project contemplates different phases that range from a review of the recent academic literature to a market study with an initial sample of 500 toys and games to assess their possibilities of raising awareness and promoting civic attitudes. As a result of this work, the CHILDTIZENS Participation Toolkit has been developed, a set of playful-educational materials aimed at promoting citizen participation and optimizing transformation from childhood, focused on 3 basic dimensions that emerged from the research: accessibility, sustainability, and gender equality.     El presente Proyecto Básico y de Ejecución de un Centro de Turismo Rural en la calle Nueva nº4 en Cuellar(Segovia). Las obras proyectadas son de promoción privada. Previa a la ejecución del edificio destinado a Centro de Turismo Rural, se realizó una toma de datos del edificio existente, para la posterior elaboración del informe de patología (en el caso de rehabilitación) y el proyecto de demolición. Se trata de un edificio de dos plantas (planta baja y primera) que data de 1930, como edificio de viviendas y locales comerciales, actualmente se encuentra en un estado deficiente de conservación, mostrando diversas lesiones (humedades, corrosión, grietas, elementos estructurales, etc.) debidas a la falta de uso y, por tanto , de mantenimiento. En la planta baja se sitúan dos locales comerciales y el portal de acceso a la planta primera. El local 1, situado en la derecha del portal, y con acceso peatonal desde la calle, se utilizaba como oficina de Caja Rural, mientras que el local 2, situado a la izquierda del portal , y con acceso rodado desde la calle, se empleó como garaje y presenta techos de doble altura y da a acceso al patio exterior. El portal está formado por un vestíbulo y una escalera de doble tramo que comunica con la planta primera. En la planta primera, se ubica un pequeño distribuidor que da acceso a dos viviendas exteriores y un pasillo que da acceso a otras dos viviendas con vistas al interior, siendo un total de cuatro viviendas y dos patios interiores. Este inmueble no se encuentra dentro del Conjunto Histórico de Cuellar, ni está catalogado como edificación protegida en ninguno de los niveles establecidos por la Normativa Urbanística, por lo tanto, no está sujeto a condiciones particulares que pudieran afectar a su proceso de demolición

    Evaluation of the immune response in betanodavirus-vaccinated gilthead seabream.

    An inactivated vaccine has been developed using a RGNNV/SJNNV reassortant strain isolated from Senegalese sole, which confers moderate protection against the homologous viral strain in sole juveniles. The aim of this study is to evaluate the differential expression of immune-related genes in gilthead seabream after vaccination, and in vaccinated fish after NNV infection. Gilthead seabream juveniles were vaccinated by intraperitoneal injection with an inactivated NNV vaccine, and control fish were injected with PBS. Thirty days after vaccination, fish were intramuscularly injected with the homologous viral strain. Head-kidney and brain samples were extracted at 2, 3 and 7 d post-vaccinaton (dpv), and at 1, 3, and 5 d post-challenge (dpc). The evaluation of the immune response was carried out using an OpenArray® platform with 56 gene targets. The vaccination triggered the differential expression of 41 of the immune-related genes analysed. All the differentially expressed genes (DEGs) showed an early down-regulation in head-kidney and/or brain, except rag1 that was up-regulated in head-kidney at 2 dpv. The il6 gene was up-regulated in the brain at 7 dpv, being the only DEG detected at this time point. After viral challenge, the response in vaccinated fish was characterized by an early up-regulation in head-kidney, with 21 out of 29 DEGs up-regulated at 1 dpc, whereas in non-vaccinated fish 15 out of 21 DEGs were down-regulated at the same time point. In brain samples from vaccinated fish, a total of 8, 36 and 32 DEGs were detected at 1, 3 and 5 dpc, respectively, all of them being up-regulated. However, a delay in the up-regulation response was observed in control fish, with most of the DEGs down-regulated at 1 dpc. In conclusion, the immune response against NNV infection in gilthead seabream seems to be modulated by the previous response to the inactivated NNV vaccine.Universidad de Málaga.     [EN] SERCC was created at the end of 2021 in response to the recent incorporation of the IGME as a National Centre of the Spanish Research Council (CSIC) and the need to converge to CSIC´s research organization, which is structured on Research Groups as basic units. SERCC is composed of 11 persons (6 Staff Scientists – 3 of them recently promoted to Scientific Researchers, 4 Specialized Technicians and 1 Predoctoral Student), and focuses on the imprint of past climate changes on the characteristics and properties of the sedimentary record, accumulated both in marine and continental realms.Peer reviewe
